What we're looking for
As a Scientist in Antibody Optimization in Cradle's growing Bioengineering team, you'll take our innovations to new heights. Your mission? To develop novel wet lab methods and workflows for antibody optimization using E. coli, S. cerevisiae, and other microbial systems.
We've reimagined experimentation with our state-of-the-art wet-lab infrastructure, seamlessly integrated with machine learning algorithms allowing for more scalable and cost-effective solutions. But that's not all – we are also developing novel techniques beyond what is mainstream!

Responsibilities
As our Antibody Optimization Scientist, you’ll:
Pave the way with cutting-edge research in antibody DNA synthesis, expression, purification, and characterization (e.g. stability and affinity)
Develop these methods into efficient, robust, and cost-effective workflows that integrate well with our BioFoundry;
Innovate on existing methods and workflows to further decrease variance, costs, turnaround time, and/or increase throughput;
Setup ultra-high throughput experiments for large-scale antibody characterization (e.g. Tite-Seq or AlphaSeq)
Utilize state-of-the-art technologies and automation to run high-throughput workflows that are fast and efficient, and generate high-quality data
Be the go-to antibody expert at Cradle, communicating your successes, results, and challenges across our interdisciplinary team
Create standard operating procedures for the innovative workflows you develop
Need-to-Haves
PhD or equivalent experience in molecular biology, biochemistry, enzymology, metabolic engineering, biophysics, or related fields.
Proven expertise in antibody expression, purification, screening and characterisation (scFvs, VHHs, etc) with in vitro and/or in vivo protein expression systems.
In-depth experience with different protein purification techniques (e.g. affinity tag purification, protein A/G chromatography) and biophysical antibody characterization techniques (SPR, BLI, DLS, ELISA, etc.)
A successful track record in antibody optimization including affinity maturation, stability and aggregation determination and functional assays
Excitement to learn, contribute, and drive innovation in an early stage startup environment. Having an appetite for its ambiguity and fast pace.
Nice-to-haves
Hands-on experience expressing antibodies in mammalian cell systems (CHO/HEK293)
Familiarity with in vitro display technologies (phage display, yeast display) involving library design, construction, and screening.
Working knowledge of structural biology and protein computational modeling approaches (e.g. Rosetta, Visual Molecular Dynamics, PyMol, RasMol, UCSF Chimera, Crystal Studio)
Experience with statistical approaches and next-gen sequencing techniques (Illumina, NanoPore, PacBio).
